Introduction: Tilia species (linden, Tiliaceae) are used in folk medicine in the world and in Türkiye for
cold, stomach ailments, and as expectorant, diuretic, sudorific, sedative, and emollient. It is also used
for cosmetic purposes due to its skin softening and hair loss preventive effects. The studies determined
that especially flowers contain mucilage, coumarin, flavonoid, triterpene, and essential oil. This study
aimed to perform isolation studies using various chromatographic methods and investigate biological
activities (anticollagenase and lipase inhibitory activity) on the methanol extract prepared from stem
bark of Tilia rubra subsp. caucasica (Rupr.) V.Engl.
Material and Method: The structures of the isolated compounds were elucidated with the help of 1H
NMR and 13C NMR spectral techniques. Anticollagenase activity was assigned spectrophotometrically
by the method using FALPGA as substrate. Lipase inhibitory activity was determined by the method
using p-nitro phenyl butyrate (p-NPB) as the substrate.
Results: As a result of the isolation studies, the main compound obtained from the methanol extract was
determined as epicatechin. The IC50 values for anticollagenase and lipase inhibitory activities of the
extract have been found as 78.242±1.241 and 81.066±1.803 µg/mL, respectively.
Conclusion and Discussion: Although further studies are needed, T. rubra subsp. caucasica may be a
promising resource for evaluation as dermocosmetic. In addition, it has significant lipase inhibitory
activity.
