Investigation of anti-cancer activity of newindole-based sulfonamide compounds indifferent types of cancer and healthy cell lines

Abstract

Rationale: Historically, cancer is a disease whose definition datesback to ancient times, but in recent years it has emerged not onlyas a global health issue but also as a socio-cultural and economicproblem. The aim of this study is to investigate the effects ofnew indole-based sulfonamide compounds, designed to be moreselective as alternatives to cytotoxic agents used in cancer treat-ments, on different cancer and healthy human cell lines.Methods: In this study, six human cell lines were cultured understandard conditions (37°C, 5%CO2), treated with indole-sulfon-amides and incubated for 24 h. Viability was assessed via MTTassay at 570 nm. Data were normalized, arcsin transformed, andstatistically analyzed using UNIANOVA in SPSS.Results: Although indole-sulfonamide compounds reduced viabil-ity at peak doses, they produced no statistically significant cyto-toxicity in ARPE-19 or BEAS-2B healthy cell lines (p > 0.05),with compound A67-24-2 maintaining over 50% viability even at250 lg/ml. In breast lineages, A50-19-2 emerged as the safestcandidate for MCF-10A cells, whereas A52-21-1 exhibited hightoxicity in healthy cells while failing to fully eradicate the moreresistant, estrogen receptor-positive MCF-7 cancer cells (p< 0.0001). Across the MDA-MB-231 and A549 cancer lines, adecisive cytotoxic threshold was identified at 62.5 lg/ml, beyondwhich all compounds induced a significant, dose-dependentdecline in cell survival (p < 0.0001).Conclusions: The synthesized indole-sulfonamide compoundsexhibited a favorable safety profile across healthy retinal andlung epithelial lines, with A50-19-2 specifically demonstrating thehighest biocompatibility in healthy breast cells. However, the statistically significant cytotoxic threshold identified at 62.5 lg/ml(p < 0.0001) revealed that while the compounds are non-toxic tomost healthy tissues, their current potency requires further opti-mization to effectively overcome the inherent resistance of MCF-7 and A549 cancer phenotypes.

Gerekçe: Tarihsel olarak kanser, tanımı antik çağlara kadar uzanan bir hastalıktır; ancak son yıllarda sadece küresel bir sağlık sorunu olarak değil, aynı zamanda sosyo-kültürel ve ekonomik bir sorun olarak da ortaya çıkmıştır. Bu çalışmanın amacı, kanser tedavilerinde kullanılan sitotoksik ajanlara alternatif olarak daha seçici olacak şekilde tasarlanan yeni indol bazlı sülfonamid bileşiklerinin, farklı kanser ve sağlıklı insan hücre hatları üzerindeki etkilerini araştırmaktır. Yöntemler: Bu çalışmada, altı insan hücre hattı standart koşullar altında (37°C, %5 CO₂) kültürlendi, indol-sülfonamidlerle muamele edildi ve 24 saat süreyle inkübe edildi. Canlılık, 570 nm dalga boyunda MTT testi ile değerlendirildi. Veriler normalize edildi, arcsin dönüşümü uygulandı ve SPSS’te UNIANOVA kullanılarak istatistiksel olarak analiz edildi. Sonuçlar: İndol-sülfonamid bileşikleri, en yüksek dozlarda canlılığı azaltmış olsa da, ARPE-19 veya BEAS-2B sağlıklı hücre hatlarında istatistiksel olarak anlamlı bir sitotoksisite oluşturmamıştır (p > 0,05); A67-24-2 bileşiği, 250 lg/ml'de bile %50'nin üzerinde canlılık oranını korumuştur. Meme hücre hatlarında, A50-19-2, MCF-10A hücreleri için en güvenli aday olarak öne çıkarken, A52-21-1, sağlıklı hücrelerde yüksek toksisite sergilemiş, ancak daha dirençli, östrojen reseptörü pozitif MCF-7 kanser hücrelerini tamamen yok edememiştir (p < 0,0001). MDA-MB-231 ve A549 kanser hatlarında, 62,5 lg/ml'de belirleyici bir sitotoksik eşik tespit edildi; bu eşiğin ötesinde tüm bileşikler, hücre sağkalımında doz bağımlı, anlamlı bir düşüşe neden oldu (p < 0,0001). Sonuçlar: Sentezlenen indol-sülfonamid bileşikleri, sağlıklı retina ve akciğer epitel hücre hatlarında olumlu bir güvenlik profili sergilemiştir; A50-19-2 ise özellikle sağlıklı meme hücrelerinde en yüksek biyouyumluluğu göstermiştir. Bununla birlikte, 62,5 lg/ml'de (p < 0,0001) tespit edilen istatistiksel olarak anlamlı sitotoksik eşik, bileşiklerin çoğu sağlıklı doku için toksik olmamasına rağmen, mevcut etki güçlerinin MCF-7 ve A549 kanser fenotiplerinin doğuştan gelen direncini etkili bir şekilde aşabilmesi için daha fazla optimizasyona ihtiyaç duyduğunu ortaya koymuştur.
